Recent developments in the field or transgenic techniques means that sophisticated mouse models can be now generated rapidly and with relative ease. The CTM’s mission is to assist researchers by not only providing cutting-edge transgenic services but to also advise them and if desired carry out all steps of transgenic model design and generation on collaborative basis.

Based on your project the CTM will suggest an optimal strategy (microinjection, electroporation, viral infection) and choice of reagents (naked DNA, CRISPR/Cas, viral vectors).

Design, Production & Injection (DPI)

Our DPI service includes everything from comprehensive allele design, embryo manipulation, breeding, and genotyping until the delivery of fully validated F1 animals.

What you provide

Provide us with sequences of desired modification and locus to allow us to design optimal transgenesis and genotyping strategies or contact us for support in allele design.

The type of template DNA (or alternative substrate for microinjection) will depend on the strategy chosen. In many cases, it is preferable for the CTM to order the appropriate template based on your sequence information (additional costs will be charged).

What we provide

We will decide on an optimal strategy to deliver template DNA and CRISPR/Cas9 RNP into wild type or transgenic embryos and transfer them to fosters.

We will design a genotyping approach to select candidate founders and breed the selected animals to F1.

We will genotype F1 animals and design a strategy to validate integrity of the desired genetic modification. Depending on the desired type of modification and transgenesis strategy chosen, this can be done either by PCR + Sanger-sequencing or through a custom ONT-based direct long-read sequencing assay (subject to additional charges).

For DPI-projects, we guarantee the delivery of validated transgenic F1 animals.

Our ROI service allows you to pay for each injection series. This service is suitable for users with previous experience and demands more effort from your side, as the genotyping and sequencing will be performed by you.

What you provide

In ROI projects, users will need to design and conduct their own genotyping assays.

What we provide

We will decide on an optimal strategy to deliver template DNA and RNP into wild type or transgenic embryos and transfer them to fosters.

We will inform you as soon as biopsies for genotyping your F0 animals are available to be picked up. Once you have genotyped them, we will organize the delivery of any F0 animals you selected.

Chimera Generation Service (Chi)

Despite the development of novel genome engineering tools that allow rapid generation of mice carrying desired mutations, extensive ES cell repositories (KOMP, EUCOMM, genetrap) offer the opportunity to generate useful mouse models. In order to facilitate this, CTM offers a ES cell microinjection service.

What we provide

Upon arrival at CTM the ES cells will be injected into at least 80 blastocysts of the appropriate recipient strain (these will vary depending of ES cell type).

The surviving embryos will be implanted into pseudopregnant foster mothers.

Any resulting chimeras will be turned over to the researcher.

What you provide

The researchers must order the selected clones from a repository at their own expense, and expand them to achieve a sufficient number of cells. The ES cell clone to be injected must be brought to the CTM as a live cell suspension on the day of microinjection.

ES clones originating from various repositories do not always produce germline competent chimeras, therefore CTM cannot offer any guarantees as to the number of pups or the number of positive pups that will be obtained. In order to increase chances of success, the CTM recommends the user to import and inject several clones targeting the same allele whenever possible.
